Chris Gunness warned that many of those reliant on food aid would be left empty-handed unless the agency can refresh its stockpiles of basic food supplies by Friday.
Israel sealed Gaza's crossings last week after Gazan militants fired rockets at Israeli border towns for several days.
Israel has not said when it plans to fully re-open the crossings but did allow a fuel shipment into Gaza on Tuesday.
That came after the territory's power plant was forced to shut down on Monday night due to the fuel shortage, plunging Gaza into darkness.
